关白附中多糖含量的测定

摘    要:目的采用水提醇沉法提取关白附粗多糖.方法经反复冻融、酶法与Sevag法联合脱蛋白后测定关白附中多糖质量分数.结果多糖经苯酚-硫酸显色后在490 nm处用紫外可见分光光度计进行比色测定,测定结果:关白附中多糖质量分数达4.62%,平均加样回收率为97.72%,RSD=1.96%.结论该方法实验操作简便,准确可靠,重现性较好.

Determination of Polysaccharide Content in Aconitum coreanum

Abstract:Aconitum coreanum was extracted with hot water,then the crude polysaccharide fraction was obtained by ethanol precipitation. After the freeze-thaw process,the crude polysaccharide was deproteinated by a combination of proteinase and Sevag method. The polysaccharide content in Aconitum coreanum was determined by phenol-sulfuric acid method comparing colors at 490 nm. Results showed that:the polysachardes content in Aconitum coreanum is 4.62% ,the average recovery rate is 97.72% and RSD is 1.96%. This method is simple,the result is stable and reproducible.
