维生素C及植物生长调节物质对油茶花粉萌发率的影响

摘    要:以油茶‘华硕’Camellia oleiferaHuashuo’花粉为试材,运用琼脂培养基萌发法研究维生素C(AsA)及植物生长调节物质对油茶花粉萌发率的影响。结果表明:适宜质量浓度的维生素(CAsA),吲哚乙酸(IAA),萘乙酸(NAA),2,4-二氯苯氧乙酸(2,4-D)和赤霉素(GA3)对油茶花粉萌发率具有显著促进作用,其中单因素处理AsA质量浓度为20 mg·L-1时,花粉萌发率达到最高(65.86%),比对照提高了23.80%;GA3质量浓度为10 mg·L-1时花粉萌发率最高,达74.22%;NAA和2,4-D处理花粉萌发率在质量浓度为1.0 mg·L-1时最高,分别达到61.33%和60.09%,IAA在质量浓度为5.0 mg·L-1时花粉萌发率达到最大(71.52%);适宜质量浓度的IAA,GA3和AsA配合使用能显著提高油茶花粉萌发率。根据适宜质量浓度设计3因素4水平正交实验,发现以5.0 mg·L-1 IAA + 5.0 mg·L-1GA3 + 20.0 mg·L-1AsA最优,油茶花粉萌发率达到82.91%。图3表3参13

Abstract:The effects of ascorbic acid(AsA) and plant growth regulators,such as gibberellic acid (GA3),naphthalene acetic acid (NAA),2,4-dichlorophenoxyacetic acid(2,4-D),and indoleacetic acid (IAA) on pollen germination percentage of Camellia oleifera 'Huashuo' were studied with in vitro tissue culture.Results for AsA alone showed that an optimal concentration of 20 mg·L-1 increased (P0.05) the pollen germination rate to 65.9%,which was 23.8% higher than the control (no AsA).The optimal concentration for GA3 alone was 10.0 mg·L-1 with a pollen germination rate of 74.22%(P0.05).The best germination rate was found with 1.0 mg·L-1 NAA,1.0 mg·L-1 2,4-D),and 5.0 mg·L-1 IAA respectively.However,the best results (pollen germination of 82.91%) was with a combination of 5.0 mg·L-1 IAA + 5.0 mg·L-1 GA3 + 20.0 mg·L-1 AsA.
