盾叶薯蓣同源四倍体的诱导和鉴定

摘    要:利用组织培养技术进行了盾叶薯蓣试管苗多倍体的诱导和鉴定。结果表明,秋水仙碱溶液中添加适量二甲基亚砜(DM SO)有利于提高诱变率。最高诱变率的处理为0.1%秋水仙碱 2%DM SO浸泡24 h,得到了20%的变异率。根据正交试验结果的方差分析和直观分析,最佳处理是0.1%秋水仙素 2%DM SO浸泡72 h,其诱变率超过20%。

Abstract:The experiment is carried out to induce and identify polyploid of Dioscorea zingiberensis C.H.Wright by tissue culturing.The process of inducing polyploid indicated that an appropriate amount of DMSO in colchicines solution helps elevate the inducing rate.In this experiment,the best way of inducing is to be dipped in the solution of 0.1% colchicines added 2% DMSO for 24 h,which induction rate hits 20%.But in the light of the result of variance and object analyses the optimum one is to be dipped in 0.1% colchicines added 2% DMSO for 72 h,which induction rate is likely to surpass 20%.
