小麦吸浆虫不同滞育虫态的RAPD研究

摘    要:应用44条随机引物,对麦红吸浆虫Sitodiplosis mosellana(Gehin)10个不同滞育阶段的个体进行RAPD检测,筛选出6条随机引物S1,S50,S81,S83,S1031,S1248,可以扩增出清晰稳定的多态性片断81条,相对分子质量在200bp~2000bp之间。应用UPGMA(非加权配对算术平均法)对多态性片断进行聚类分析,构建树状图,推测不同滞育阶段之间的聚类关系。结果表明:在各个滞育阶段中,裸露幼虫相互之间的遗传距离和亲缘关系比较近,具有比较同质的遗传背景,而不同阶段的结茧幼虫相互之间的关系比较复杂。

Abstract:The phylogenetic relationship of ten kinds of status that are of different diapause characters in the wheat blossom midge,Sitodiplosis mosellana(Gehin) was studied by using RAPD technique.The genomic DNA of ten samples was amplified with 44 random primers.Only six primers(S1,S50,S81,S83,S1031,S1248) could produce clear fragments and obtained a total number of 81 fragments in nine samples amplified.Molecular size of the fragments ranges from 200bp-2000bp.The phylogenetic relationship was inferred by using UPGMA based on RAPD makers.The results showed that the genetic relationship of the non-cocooned larva is similar but it is complex of the cocooned larva.
