丹参提取物体外抗氧化活性研究

摘    要:用φ=70%的乙醇超声提取丹参粉末,减压浓缩得到乙醇浸膏,加蒸馏水制成悬浮液,依次用等体积的石油醚、氯仿、乙酸乙酯、正丁醇萃取,得到不同溶剂萃取物。通过体外抗氧化试验,研究丹参不同溶剂萃取物清除DPPH.、H2O2、.OH和还原Fe3+的能力(以BHT为对照)。结果表明,丹参4种提取物均具有不同程度抗氧化活性,且抗氧化活性与提取物质量浓度呈量效关系。乙酸乙酯提取物清除H2O2、.OH的能力均较强,IC50分别为0.513和0.650 g/L;正丁醇提取物清除DPPH.能力最强,IC50为0.059 g/L。丹参提取物具有较强的抗氧化活性,可作为一种新的自由基清除剂和天然抗氧化剂。

Abstract:The antioxidant activities of the different polar solvents root extracts of Salvia miltiorrhiza Bunge.were studied to provide a basis for its further development and utilization by antioxidant test in vitro.The powdered roots of Salvia miltiorrhiza Bunge.were extracted with 70% ethanol ultrasonic extraction,ethanol condensate were obtained under reduced pressure and were evaporated to dryness under vacuum on a rotary evaporator.The dry condensate was dissolved in water,the made water pending liquid was obtained and extracted different solvents with the same volume of petroleum ether,chloroform,ethyl acetate,n-butanol in turn.Its antioxidation activities were detected and evaluated by four methods of DPPH,H2O2,·OH,and reducing power of Fe3+(for comparison BHT).The results indicated that four kinds of solvent extracts of Salvia miltiorrhiza Bunge.have a certain degree of antioxidant capacity in four determination system.And it also showed that the extract concentration was higher and the effect better.The ethyl acetate extraction had better effect on scavenging H2O2,·OH,and reducing power,and the IC50 was 0.513 g/L,0.650 g/L respectively.The n-butanol extraction had better effect on scavenging DPPH radical,and the IC50 was 0.059 g/L.The ethyl acetate extraction of Salvia miltiorrhiza Bunge.has strong antioxidant activity,which can be used as a new free radical scavenging scavenger and natural antioxidants.
