山羊子宫内膜腺上皮细胞的分离培养和鉴定

摘    要:为了探讨山羊子宫内膜腺上皮细胞的分离、生长条件以及纯度鉴定,通过2 g/L的Ⅰ型胶原酶置于水浴消化4 h,200目的滤网过滤后进行低速离心,收集沉淀物,沉降洗涤3~5次,将细胞移至24孔培养板置37℃细胞培养箱中进行培养。12 h后开始贴壁生长,腺团开始向外部扩散,生长2~3 d,细胞扩散并出现明显的铺路石状,4~5 d腺团基本扩散开来并呈一定走向继续生长。经免疫组织化学方法进行细胞染色,细胞表达角蛋白的阳性率达90%以上。
