杜梨果实多糖提取方法及含量测定的研究

摘    要:分别采用超声波-微波协同萃取、微波、水浴及超声波4种方法提取杜梨果实多糖,用蒽酮-硫酸比色法测定多糖的含量。结果表明,4种提取方法存在明显的差异,以超声波-微波协同萃取效果最佳,其后依次是微波、水浴及超声波。4种方法提取下多糖的含量分别为13.91%、13.56%、12.74%和11.06%,葡萄糖浓度在25.15~100.6μg.mL-1范围内呈现良好的线性关系,平均回收率为100.5%,RSD为1.59%(n=5)。超声-微波协同萃取提取杜梨果实多糖的方法优于微波、水浴及超声波法提取,蒽酮-硫酸比色法测定多糖含量的方法准确,重复性好。

Abstract:Ultrasonic-microwave synergistic,microwave,water bath and ultrasonic extraction were employed to extract the polysaccharide and ant hrone-sulphuric acid colorimetry was used to determine its content.There exist remarkably differences among the four methods;ultrasoni-cmicro wave synergistic extraction is the optimal method,then the microwave,water bath and ultrasonic extraction following it.The polysaccharide content of the four methods is 13.91%,13.56%,12.74% and 11.06% respectively.The linear range was 25.15~100.6 g/mL-1 for glucose.The average recovery rates 100.5%(RSD 1.59%,n=5).The ultrasonic-microwave synergistic extraction is the best extraction method.The method of anthrone-sulphuric acid colorimetry determine the polysaccharide content was stable,accurate.
