| 摘 要： | 为从核桃叶片中提取高质量的基因组总DNA,比较了9种DNA提取方法。结果表明,改良的CTAB法9提取的DNA溶液无色透明,紫外分光光度计测得A260/A280为1.8~2.0,用0.8%琼脂糖胶电泳,为一条清晰的带,蛋白、多糖、酚类物质、RNA等去除较彻底,DNA无降解现象,该DNA能够适应AFLP分析的要求。

| Abstract: | To get high-quality DNA from leaves of walnut,nine different DNA-extracting methods were compared in this paper.The result indicated that the improved CTAB 9 appeared as a clear band,and then electrophoresed in 0.8% agarose gel,the polyhexose,protein and RNA were eliminated completely.Measured the DNA by smartspec TM 3 000,the value of A_(260) /A_(280) was 1.8~2.0.Therefore,this method is completely suitable for AFLP. |
