重寄生链霉菌F46与链霉菌SC1融合子的筛选

摘    要:运用原生质体融合技术将重寄生链霉菌F46与生防链霉菌SC1融合,获得40株融合子.依据形态特征、皿内拮抗试验对获得的融合子进行初筛,发酵液抑菌活性测定,筛选出2株抑制效果明显的融合子FSf-11和FSf-13.皿内拮抗试验结果表明,2个融合子对灰葡萄孢(B.cinerea)有明显的抑制作用,抑制率比亲本SC1分别提高了27.99%和20.56%,镜检发现灰葡萄孢的基内菌丝畸形,经FSf-13处理后还出现原生质凝聚现象.融合子的发酵滤液对胶孢炭疽菌(C.gloeosporioides)抑制效果显著,抑菌圈直径超过了20 mm;融合子FSf-11的发酵滤液对尖镰孢萎蔫专化型(F.oxysporum f.sp.vasinfectum)和大丽轮枝菌(V.dahliae)的孢子萌发抑制作用最强,其孢子萌发率低于10%;融合子FSf-13对2种靶标菌的孢子萌发抑制率也超过了50%.

Abstract:Forty fusants were obtained from protoplasts fusion with hyperparasitic Streptomyces F46 and Streptomyces SC1 and two fusant FSf-11 and FSf-13 were screened basing on the morphological characteristics and antibiotic activity in vitro. Evident inhibiting activity against Botrytis cinerea were found and inhibiting rate of fusant FSf-11 and FSf-13 to B. cinerea increased 27. 99% and 20. 56% comparing with the strain SC1. Malformation of substrate mycelium and protoplast coacervation in mycelium of B. cinerea were observed with microscope. Fermentation filtrate of the fusants had evidently inhibiting effect on mycelium growth of C. gloeosporiaides and conidia germination of F. o. f. sp. vasinfectum and V. dahliae.
