摘    要:以黄瓜未受精子房为试材,研究了激素种类、暗培养、温度等对启动雌核发育和胚状体诱导的影响,旨在探讨建立黄瓜未受精子房培养获得再生植株的技术体系.结果表明,未授精子房预培养阶段,25℃下暗培养1 d,启动雌核发育的反应率最高可达91.3%;TDZ浓度对启动雌核发育的影响与温度有关:相同TDZ浓度下,25℃比35℃更适合启动雌核发育;25℃条件下,启动雌核发育的最适TDZ浓度为0.02 mg/L,反应率可达90.1%;35℃热激处理中,启动雌核发育的最适TDZ浓度为0.06 mg/L,反应率为61.1%;胚状体诱导阶段,培养基中添加1.0 mg/L6-BA时,平均每子房产胚量最高,为10.53个.

Abstract:The aim of this research was to establish a plants regeneration system for cucumber(Cucumis sativus L.) via in vitro gynogensis in order to provide a quick way to obtain self-bred lines for cucumber F1 breeding.It researched the effects of the growth regulators,dark culture and temperature on induction of gynogenesis and embryoids.Data indicated that in preliminary culture the maximum frequency of gynogenesis response was 91.3% in 25 for 1 day dark culture.25 was better for the initiation of genogensis compared with 35 in medium supplemented the same concent ration of TDZ.The maximum frequency of gynogenesis response was up to 90.1% in CBM medium supplemented 0.02 mg/LTDZ in 25 treatment,while this frequency was 61.1% in CBM medium supplemented 0.06mg/LTDZ in 35 treatment.The maximum number of embryoid induction was 10.53 perovary in CBM medium supplemented 1.0 mg/LBAP.
