布氏杆菌单克隆抗独特型抗体细胞株建立及免疫原性测定

摘    要:在建立了布氏杆菌单克隆抗体细胞株的基础上,筛选具有强保护作用的单抗细胞株A7免疫BALB/C鼠,并进行细胞融合.用竞争抑制ELISA法筛选出5个强阳性孔,经克隆化培养后首次建立了9个布氏杆菌单克隆抗独特型抗体细胞株.其中,F9株传25代后及液氮冻存复苏后仍能稳定地产生高效价的抗独特型抗体.其IgG类型鉴定为IgG2a,并证实具有抗原“内影象”.以鼠红细胞为载体将抗独特型抗体吸附其表面进行小鼠免疫试验,检测结果表明该抗独特型抗体具有良好的免疫原性,能诱导免疫小鼠产生相当效价抗布氏杆菌抗体(Ab3).

Abstract:After the establishment of Mcabs to a local brucella strain S85A,the McAb A7 was selected and tested as a protective one.Using McAb A7 to immunize BALB/c mice in order that cell fusion was carried out.Five positive wells were selected out by competitive inhibiting ELISA.After cloning culture we established 9 monoclone anti-idiotypic cell strains.F9, one of them, could prodace higher titer anti-idiotypic Ab.than others did and could do so stably after 25 generations cuture and frozen-thaw procedure.And this anti-idiotypic Ab.was proved to possess "internal image" of S35A while its IgG subtype was identified as IgG2a.The test of immunizing BALB/c mice was conducted by using the red cell of mice as a carrier to conjugate the anti-idiotypic Ab.to it.Those mice had produced certain titer Ab to S35A (A63).This result suggests that the antiidiotypic Ab.F9 not only possess the Ag "internal image",but have the available immunogenicity to stimulate mice producing Ab3.
