壳寡糖诱导油菜抗菌核病机理研究初探

摘    要:壳寡糖作为一种有效的生物农药已被应用于多种农作物,田间使用发现其可诱导油菜抗油菜菌核病,但机理不甚明了。本试验证明此诱抗有时间依赖性,接种核盘菌前提前3 d用50μg/mL浓度壳寡糖预处理的植株有最佳防治效果,防效高达72.1%。而平板抑菌试验证明壳寡糖对核盘菌的生长没有直接抑制作用,说明油菜对菌核病的抗性来源于壳寡糖激发的植物自身系统抗性。利用半定量RT-PCR检测发现油菜中重要的抗性基因BnPDF1.2可被壳寡糖诱导表达水平升高,壳寡糖还可以诱导茉莉酸生成途径中关键酶脂氧合酶(LOX)的活性升高,说明壳寡糖诱导油菜抗菌核病可能由JA/ET途径介导。

Abstract:Oligochitosan was considered to be a potent plant defense elicitor.In this paper the inducing resistance of oligochitosan to Sclerotinia sclerotiorum(SS) on Brassica napus was studied. Whereas oligochitosan did not affect radial growth of SS colonies on potato dextrose agar plates, it reduced the frequency and size of rot compared to untreated controls when applied to oilseed rapes before inoculation with SS. The best inducing resistance concentration of oligochitosan was 50 Lg·mL-1 nd the best pretreated time was 3 days before inoculation with SS. mRNA levels of PDF 1.2 were induced by oligochitosan, and the activity of a jasmonate acid biosynthesis enzyme lipoxygenase(LOX) was induced by oligochitosan on leaves. These results suggest that oligochitosan treatments induced Brassi2 canapus host resistance to S. sclerotiorum through the JA/ET signaling pathway.
