| 摘 要： | 以六月酥梨叶片为外植体,研究了基本培养基、不同质量浓度BA和IBA的组合、不同暗培养时间、不同碳源种类和质量浓度、叶片放置方式和乙烯抑制剂AgNO3处理等因素对六月酥梨叶片再生体系的影响。结果表明,六月酥梨叶片再生芽的适宜组合为NN69+BA 5.0 mg/L+IBA 0.3 mg/L+山梨醇40 g/L+琼脂7 g/L,远轴面向下,暗培养30 d。在适宜条件下,六月酥梨叶片再生频率高达83.3%,平均再生芽数为2.84。不同碳源种类和质量浓度对六月酥梨叶片再生有不同的影响,其中40 g/L山梨醇对叶片不定芽诱导效果显著。硝酸银质量浓度为0.1~1.0 mg/L时六月酥梨叶片再生频率高于对照,但无显著差异;当硝酸银质量浓度为2.0 mg/L时,显著降低了叶片再生频率。叶片再生苗在生根培养基1/2MS+IBA 0.5 mg/L+蔗糖20 g/L+琼脂6.5 g/L,暗培养5 d,生根率为50%,平均生根数为3.33。
