白菜型冬油菜 ELIP1基因的克隆与低温表达分析

摘    要:为探讨 ELIP1基因在冬油菜抗寒中的作用,克隆白菜型冬油菜 ELIP1基因的CDS序列,并对其功能结构域、系统进化和不同抗寒品种间在低温下的表达特性进行分析。结果表明,ELIP1蛋白编码195个氨基酸,预测分子质量为20.472 8 ku,等电点9.24。序列比对分析表明, ELIP1基因在弱抗寒品种发生突变位置少于抗寒品种;进化树分析结果显示,弱抗寒品种‘Lenox’和‘Neib’聚为一类,‘天油2号’‘天油4号’‘陇油6号’‘陇油7号’‘陇油8号’和‘陇油9号’聚为一大类。qRT-PCR分析表明,冬油菜 ELIP1基因在不同抗寒品种间存在表达差异,低温下, ELIP1基因在超强抗寒品种中表达水平显著高于抗寒性相对较弱的品种,说明 ELIP1基因在冬油菜响应冷胁迫中具有重要作用。

Abstract:The CDS sequence of ELIP1 gene was cloned from Brassica rapa L. and its functional domain, phylogenetic evolution, gene structure and its expression under cold stress were analyzed to study the role of ELIP1 gene in cold tolerance . The results showed that the cDNA sequence of this gene encoded a protein of 195 amino acid residues, which a predicted molecular mass was 20.472 8 ku and a theoretical pI value was 9.24. Sequence alignment demonstrated that the mutation position of ELIP1 gene in poor cold-tolerant cultivars was less than that in strong cold-tolerant cultivars. Phylogenetic tree analysis showed that poor cold-tolerant cultivars Lenox and Neib belonged to the same class, and other cultivars were clustered into a large category. QRT-PCR analysis revealed that the expression of ELIP1 gene was different in different cold-tolerant winter rape cultivars. ELIP1 gene expression levels in strong cold-tolerant cultivars were significantly higher than in poor cold-tolerant cultivars under low temperature. These results showed that ELIP1 gene plays an important role in response to cold stress of winter rape.
