糯玉米DNA提取方法的优化及SSAP方法的建立

摘    要:在综合现有DNA提取方法和试验材料本身性质的基础上,对糯玉米基因组DNA提取方法进行了优化和改进,筛选出一种适合于糯玉米特异序列扩增多态性(sequence-specific amplification polymorphisms,SSAP)分析的高纯度DNA提取方法。所得DNA的A260/280在1.8~2.0间,A260/230大于2.0,琼脂糖凝胶电泳检测结果也证明该方法获得的DNA完整、无降解现象。通过对不同酶切组合、引物组合等因素的对比试验,初步建立了适用于糯玉米SSAP分析的方法体系,得到了一组条带清晰和多态性丰富的图谱。

Abstract:Base on the existed DNA extraction methods and the nature of waxy corn,waxy corn genome DNA extraction method has been optimized and improved.A260/280 of the DNA obtained by the method is between 1.8 and 2.0,and A260/230 is more than 2.0.The test result of agarose gel electrophoresis have proved that the DNA is compiete.it is more suitable for sequencespecific amplification polymorphisms(SSAP) analysis.Through comparative experiments,such as the combination of different enzymes restriction,different primers combination,SSAP analysis has been established on waxy corn,Clear bands and high polymorphisms have been got.The research made a foundation of germplasm,genetic diversity and fine breeding for waxy corn.
