dw基因mRNA在矮小型优质肉鸡S2系 不同组织的表达差异分析

摘    要:以矮小型优质肉鸡品系(S2系)为素材,采用RT-PCR技术分析dw基因在12周龄S2鸡肝脏、胸肌、腿肌和骨膜中等组织中的表达情况.结果表明,dw基因在肝脏、胸肌、腿肌和骨膜中均有表达,但在不同组织中的表达量不同.公母鸡在肝脏的mRNA水平(公鸡拷贝数为11.2±1.11,母鸡拷贝数为10.4±0.51)均显著高于胸肌、腿肌和骨膜(P<0.05);公母鸡在骨膜中的mRNA表达量(公鸡拷贝数1.8±0.37,母鸡拷贝数1.6±0.4)最低,且显著低于肝脏、胸肌和腿肌(P<0.05);母鸡在腿肌中的表达量(拷贝数为8.0±0.32)显著高于胸肌中的表达量(拷贝数为4.0±0.32)(P<0.05);公鸡在腿肌和胸肌中的表达量差异不显著.dw基因mRNA在肝脏和骨膜中的表达无性别效应,在胸肌和腿肌中的表达有明显的性别效应.

Abstract:S2 line of dwarf chicken, as quality broiler, was caused by the sex-linked dwarfing (SLD) gene. In order to illustrate the influence of dw gene on the growth performance, the expression patterns of dw mRNA were examined in four growth related tissues (liver, breast muscle, thigh muscle, periosteum) at 12-week-old female and male of S2 line of SLD chicken. Results of the real time-PCR showed that dw gene was expressed in liver, breast muscle, thigh muscle and periosteum, but its expression abundance in different tissues was different. dw gene mRNA for highest level expression was in the liver of male chicken and female chicken(copy number of male chicken was 11.2±1.11 and female chicken was 10.4±0.51), and was significantly higher than that in its breast, thigh muscle and periosteum(P<0.05). The mRNA level of dw gene in the periosteum of S2 line of SLD chicken(copy number of male chicken was 1.8±0.37 and female chicken was 1.6±0.4) was significantly lower than that in liver, breast and thigh muscles(P<0.05). dw gene mRNA for highest level expression in thigh muscle was significant than that in female chicken (P<0.05).There is no obvious different between the mRNA level of dw gene in breast and thigh muscles of male chicken (P>0.05). The expression level of dw gene mRNA in liver and periosteum of female chicken has no obvious sex effects, but in breast and thigh muscles has extremely significant sex effects.
