4个绵羊品种PTGS2基因部分片段的多态性分析

设计2对引物,采用PCR-SSCP和PCR-RFLP技术检测前列腺素内过氧化物合酶2(PTGS2)基因在小尾寒羊、湖羊、多赛特羊、萨福克羊4个绵羊品种304个个体中的单核苷酸多态性.结果表明,PTGS2基因引物1扩增片段在4个绵羊品种中不存在PCR-SSCP多态性.引物2扩增片段存在PCR-RFLP多态性,在4个绵羊品种中都检测到2种基因型(AA、AB),都没有检测到BB基因型.A等位基因频率明显高于B等位基因频率.小尾寒羊与多赛特羊、萨福克羊、湖羊之间PTGS2基因型分布差异显著(P＜0.05),其余品种之间PTGS2基因型分布差异都不显著(P＞0.05).

Polymorphism analysis of partial fragment of PTGS2 gene in four sheep breeds

The single nucleotide polymorphism of prostaglandinendoperoxide synthase 2(PTGS2)gene was detected in four sheep breeds(Small Tail Han,Hu,Dorset and Suffolk) by PCR-SSCP and PCR-RFLP with two pairs of primers.The results indicated that there was no PCR-SSCP polymorphism in four sheep breeds detected by primer 1.There was PCR-RFLP polymorphism in four sheep breeds detected by primer 2.Two genotypes(AA and AB) were detected in four sheep breeds and BB genotype was not detected.Frequency of allele A was obviously higher than that of allele B.The PTGS2 genotype distributions were significantly different(P<0.05) among Small Tail Han sheep and Dorset,Suffolk,Hu sheep.
