褐飞虱不同生物型基因组DNA的RAPD分析

摘    要:试验了两种DNA提取方法抽提褐飞虱基因组DNA的效果及其用所获得的DNA进行RAPD扩增的技术条件,结果发现,采用蚜虫DNA的提取方法(略有改进)来抽提高飞虱基因组DNA,并用S系列的随机引物、RAPD的通用反应体系及94℃变性,36℃退火和72℃延伸的扩增条件来对其进行随机扩增,可获得满意的效果,不同致害类群的群体间和同一致富类群的个体间的RAPD多态性均有差异,单雌纯化只能将个体间的差异缩小,

Abstract:A procedure of randomly amplified polymorphism DNA (RAPD) for analyzing genome DNA of brown planthopper (BPH), including the extraction method of DNA, amplifying reaction system and conditions etc was provided in this paper Moreover, the RAPD analysis on genome DNA of different BPH biotypes was conducted The results showed that there existed significant RAPD difference between various biotypes and among individual BPHs, and genetic purifying could lessen this difference in the level of individuals but not in the level of biotypes
