摘    要:将猪瘟病毒(Hog Cholera Virus,HCV)E2蛋白用大肠杆菌表达纯化后用作弱毒抗原;猪瘟病毒(HCV)强毒株用PK-15细胞培养48h,破碎后进行蔗糖密度梯度离心,吸取30%~35%梯度之间的条带,纯化后用作强毒抗原,然后用胶体金标记抗原,运用双抗原夹心法于国内首先建立了HCV抗体检测的免疫层析试纸条。该方法操作简单、灵敏度高、特异性好,能区分强弱毒感染,适合猪瘟的临床诊断。

Abstract:Protein E2 of Hog Cholera Virus was expressed in E. coll. After purified, the gene- expressed E2 acts as attenuated HCV antigen. The PK- 15 cells infected by HCV titration of virulent strain were cultivated for 48 hours. Then the PK- 15 cells were braked and performed a sucrose density gradient centrifugation. The proteins between 30% -- 35% gradient were purified and served as virulent HCV antigen. The HCV antigens were labeled by colloidal gold. Based - on sandwich ELISA technique,immune chromatographic assay test paper for detecting HCV is firstly developed in China. The method can discriminate between virulent and attenuated strain, which is more sensitive and specific than other methods and fit for clinical diagnosis of HCV.
