马铃薯试管薯诱导因素研究

摘    要:以马铃薯栽培品种克新3号脱毒试管苗为材料进行试管薯诱导技术研究。结果表明,全黑暗条件对试管薯形成、结薯数量和平均鲜重均有极显著的促进作用;适当低温(19±1)℃有利于试管薯的形成和膨大;液体培养基诱导试管薯的效果优于固体和固液双层培养基,是生产试管薯最好的培养方式;培养基(MS+8.0%蔗糖)中添加100mg

Abstract:The virus-free tube shoots of the potato cultivar Kexin 3 wereused as the materials in the study.The results showed that the formation,amount and average fresh weight of the tube miniature potatoes were significantly increased in full-darkness.The moderate low-temperature(19±1)℃ was advantageous in forming and inflating the tube miniature potatoes.The inductive efficiency of the liquid medium was higher than those of the solid medium and the double layer medium,liquid medium was the best one to product the tube miniature potatoes.The medium(MS+Sugar 8.0%) being added CCC 100 mg·L-1 could induce the formation of the tube miniature potatoes and significantly increase its amount and the yield of fresh tubers.And the medium with CCC 500 mg·L-1 +6-BA 5 mg·L-1 could significantly increase the rate of big tubers and the weight of fresh tubers,but the number of growing tubers was significantly decreased.The inductive effect of the medium only with 6-BA 5 mg·L-1 was low.It showed that the kinin 6-BA was not an essential factor for inducing tubers in plant tissue culture.
