百合上黄瓜花叶病毒的一步RT-PCR检测

摘    要:根据黄瓜花叶病毒的外壳蛋白基因序列设计、合成引物,采用Trizol快速提取百合感病组织和健康组织的总RNA,运用RT-PCR两步法和一步法都可从感病组织中扩增出与预期的335 bp大小一致的目标片段,而健康组织无此扩增产物。两种方法相比,一步RT-PCR法特异性强、灵敏度高、省时经济、程序简单,达到了快速检测的目的。

Abstract:A pair of specific primers were designed and synthesized based on the nucleotide sequence of the coat protein gene of cucumber mosaic virus(CMV),and Trizol was adopted to extract the total RNA from the virus-infected tissue and the healthy one.The expected product(335 bp) could be amplified by two-step RT-PCR and one-step RT-PCR from the infected samples,while no amplified products were obtained from the healthy tissue samples.Between the two methods,one-step RT-PCR was more special,sensitive than two-step RT-PCR.It reaches the aim of quick detection.
