摘    要:用转基因耐盐玫瑰的硬枝带芽茎段、绿枝带芽茎段、嫩叶为外植体进行组织培养快速繁殖研究,通过诱导培养,从中选出适于组织培养的外植体。试验表明:绿枝带芽茎段是转基因耐盐玫瑰组织培养的最佳外植体;诱导芽生长的最适培养基为MS+6-BA 0.5 mg/L,诱导率为26.67%;诱导芽增殖的最适培养基为MS+6-BA 0.5 mg/L+ZT 0.5 mg/L,增殖倍数为8;诱导生根的最适培养基为1/2 MS+NAA 0.5 mg/L,每苗生根数为11;移栽以河砂∶蛭石∶草炭=1∶1∶1的栽培基质为最佳,成活率达85%以上。

Abstract:Hard branches with buds, greenwoods with buds and leaves explants of salt tolerance Rosa rugosa var. of transgenosis were cultured on Murashige and Skoog (MS) basal medium supplemented with different concentrations of either 6-BA solely or in combination with naphthalene-3-acetic acid (NAA). The best response in terms of the percentage of shoot regeneration was obtained from greenwoods with buds cultured on MS medium supplemented with 0.5 mg/L 6-BA, whereas'the highest number of shoots per responding explant was recorded on medium containing 0.5 mg/L 6-BA and 0.5 mg/L ZT. Successful' rooting was achieved by placing the microshoots onto 1/2 MS medium containing 0.5 mg/L NAA. Of the various substrates tested, riversand " vermiculite " peat= 1 : 1 " 1 was the best for plant acclimatization, as 85% of the plants survived.
