摘    要:在不同营养条件以及培养环境条件下,测试了甘薯黑斑病菌的生长速度以及产孢量。结果表明:甘薯黑斑病菌(Ceratocystis fimbriataEllis et Halsted)在13种供试培养基上均能生长,其中在番茄培养基上生长最好,在胡萝卜培养基上生长最差;分生孢子产孢量以在PDA培养基上最多,在玉米粉培养基上最少。菌丝适宜生长温度为25℃,在15℃下分生孢子产孢量最大。在全光照环境下生长最好,全黑暗下次之,光暗交替条件下较差,光照条件对黑斑病菌分生孢子产孢量没有明显差别。菌丝适宜生长的pH值范围为6~8,最适pH值为8,分生孢子最适产孢pH值为10。在真菌生理培养基上,以淀粉为最佳碳源,以酵母膏为最佳氮源。菌丝、分生孢子和子囊孢子的致死条件分别为52℃、5 min,47℃、5 min和50℃、5 min。

Abstract:The effects of culture conditions on the growth and sporulation quantity of Ceratocystis fimbriata were studied.The results showed that the pathogen could grow on 13 different kinds of culture media.The best medium for growth was tomato medium,while carrot medium was the worst one.The optimal condition for spores germination was corn meal agar,while PCA was the worst one.The optimum temperature for mycelium growth was 25 ℃,at 15 ℃ the sporulation quantity was the largest.The mycelium grew best under full illumination,then under total darkness,and that with alternation of light and darkness was the worst.There was no obvious difference in sporulation quantity under different light conditions.The suitable pH value for mycelium growth was 6-8,the optimum was 8,and that for sporulation was 10.Starch and yeast extract were the best carbon source and nitrogen source,respectively,for the pathogen growth on the fungi physiological medium.The lethal temperatures of mycelium,conidia and ascospore were 52 ℃ for 5 min,47 ℃ for 5 min and 50 ℃ for 5 min.
