卡那霉素筛选转基因小麦种子关键技术研究

摘    要:对影响小麦卡那霉素筛选效果的培养基质和药剂处理方式进行了比较研究.结果表明:在种子吸胀阶段进行充分有效的卡那霉素浸种处理是提高卡那霉素筛选效果的关键技术;种子不经卡那霉素溶液浸种吸胀而直接在蛭石基质上,或用水浸种吸胀后再在蛭石等培养基质上进行卡那霉素处理,会降低小麦卡那霉素抗性筛选的效果.小麦种子幼苗的卡那霉素筛选,可用100 mg/L卡那霉素浸种至露白后再放到装有蛭石的培养皿上浇水即可,不需要再浇卡那霉素溶液.

Abstract:Comparative studies indicated that the most important step for effective transgenic wheat screening,using kanamycin as select chemicals and wheat seed as screening target,was to put the dry wheat seeds into kanamycin solution at a concentration of 100 mg/L for about 24 to 48 h,until the wheat seeds were suffiently swollen and the whitish coleoptiles were visible.The effect would be reduced greatly if the seeds were not saturated in kanamycin solution,or were saturated in water,before their transfer onto vermiculite medium,even if they were further watered with kanamycin solution after the transfer.An economic and effective practical protocal was present,which was characterized by saturating dry wheat seeds with kanamycin solution until the whitish coleoptile visible and then transfering onto vermiculite and being watered with solution without kanamycin chemicals or with only pure water.
