摘    要:不同植物花粉的自身特性不同,导致不同的染色方法只适合某些植物花粉生活力的测定。以能够在河北省石家庄地区自然开花结实的甘薯品种河北351及其不同的自交后代材料为试材,利用目前常用的4种花粉染色法(I-KI染色法、TTC染色法、过氧化物酶染色法和蓝墨水染色法)分别测定甘薯花粉的生活力,以筛选出适宜的甘薯花粉生活力测定方法;并利用该方法,对河北351自交各世代群体中自然开花株系的花粉生活力进行检测,以验证方法的可行性以及检测结果的可靠性。结果表明:适宜甘薯花粉生活力检测方法为TTC染色法,且方法可行、结果可靠。该方法测定甘薯花粉生活力具有操作简便快捷、成本低、适合大批量检测等优点,可为开展甘薯亲和性相关研究奠定基础。

Abstract:The sweet potato variety Hebei 351 and its self-crossed progeny were used as the materials,which was naturally flowering and fruiting in Shijiazhuang.The pollens were stained by four methods(I-KI method,TTC method,peroxide enzyme method and blue-ink method)to screen the suitable one.The feasibility and reliability were tested.The results showed that TTC method was suitable for sweet potato with reliability as well as feasibility.The method had some advantages such as simple,rapid,low cost and suitable for mass serial detection and so on,which laid a foundation for further study on the compatibility of sweet potato.
