原花青素对Aβ25—35诱导PC12细胞氧化应激反应的保护作用

摘    要:目的探讨原花青素对β-淀粉样肽25-35(Aβ25-35)诱导PCI2细胞凋亡及氧化应激反应的影响。方法3×10^8/LPCI2细胞培养24h,分别加入培养液(对照组)、10μmol/LAβ25-35(诱导组)、30mg/L原花青素+10μmol/LAβ25-35(保护组)。24h后收集细胞,以Hoechst33258荧光染色观察细胞凋亡,比色分析测定MDA、H2O2含量和CAT活性。结果诱导组细胞可见凋亡特异性核固缩、核碎裂;MDA和H2O2含量与对照组比较明显增加(P〈0.01),CAT活性明显降低(P〈0.05)。保护组与诱导组比较,凋亡特异性核固缩、核碎裂减少;MDA和H2O2含量明显降低(P〈0.01),而CAT活性显著增强(P〈0.01)。结论原花青素可抑制Aβ25-35诱导的PC12细胞凋亡,可能与其抑制Aβ25-35引起的氧化应激反应有关。

Abstract:Objective To study the effect of procyanidine on β -amyloid peptide 25-35 (A β 25-35) induced apoptosis and oxidative stress reaction in PC12 ceils. Methods PC12 cells with a concentration of 3 ×10^8/L were cultured for 24 h, and then incubated with culture solution (control group), 10 μmol/L A β15-35 (induction group), and 30 mg/L procyanidine plus 10μmol/L Aβ 25-35 (protection- group). Cells were harvested 24h later, and their apoptosis, MDA and H2O2 levels, and CAT activity were determined by Hoechst 33258 fluorescence staining and colorimetric analysis, respectively. ResuLts In induction group, apoptosis-specific pyknosis and kary-orrhexis were observed, MDA and H2O2 contents increased (P〈0.01), but CAT activity decreased (P〈0.05) compared with controI group. However, compared with induction group, apoptosis-specific pyknosis and karyorrhexis and MDA and H2O2 levels decreased (P〈0.01), but CAT activity increased (P〈0.01) in protection group. Conclusion Procyanidine can inhibit A β25-35 induced apoptosis of PC12 cells, which may be related to the suppression of oxidative stress reaction.
