双氧水对鼻咽癌细胞生长特性的影响及其机制的初步研究

摘    要:目的:观察H2O2对人低分化鼻咽癌细胞(CNE-2Z)生长、增殖能力的影响,并探讨其可能机制。方法:用M TT及平板克隆形成实验、W estern B lot等检测H2O2诱导CNE-2Z后增殖能力、EGR-1、p53、p16、Cyclin D1的表达。结果:不同浓度H2O2可不同程度地抑制CNE-2Z细胞的生长增殖能力,同一时间点各实验组细胞抑制率相比差异有统计学意义,抑制率与H2O2浓度间表现出明显的剂量-效应关系。平板克隆形成实验的抑制率较M TT更明显。CNE-2Z无EGR-1、p16蛋白表达,可见p53、Cyclin D1蛋白表达;诱导培养后EGR-1、p53、Cyclin D1蛋白表达明显增加。结论:一定浓度的H2O2能抑制CNE-2Z细胞生长,其作用可能与EGR-1及p53蛋白表达增强有关;Egr-1基因在CNE-2Z中具有可诱导性,在肿瘤治疗中有一定的应用前景。

Abstract:Objective: To observe the effects of H_2O_2 on the proliferation of nasopharyngeal carcinoma cell line(CNE-2Z) and study the possible mechanisms.Methods: The CNE-2Z cells were treated with H_2O_2;the assays of MTT and plate colony formation were used to measure the ability of proliferation of CNE-2Z cells;the western blot was used to detect the protein levels of Egr-1,p53,p16 and cyclin D1.Results: In a dosage dependent manner,H_2O_2 inhibited the proliferation of CNE-2Z cells,shown by both the MTT and plate colony formation assays with stronger displaying in the plate colony formation assay;without treatment of H_2O_2 CNE-2Z cells express slight p53 and cyclin D_1 proteins but no detectable Egr-1 and p16 proteins;after treatment with H_2O_2,CNE-2Z express significant higher proteins of Egr-1,p53 and cyclin D1 with no detectable change p16 protein.Conclusion: H_2O_2 might inhibit the proliferation of CNE-2Z through increasing the expressions of Egr-1 and p53 proteins;since Egr-1 expression was inducible,Egr-1 might be a target in the tumor therapy.
