摘    要:目的探讨高良姜二苯基庚烷类、黄酮类、挥发油类及高良姜醇提物(组分Ⅰ-Ⅳ)对人肝癌HepG2细胞过氧化损伤的保护作用。方法采用减压硅胶柱层析(VLC)法对高良姜提取物进行分离;采用薄层色谱法(TLC)结合高效液相-质谱联用(HPLC-MS)法对各组分进行归属与鉴定;建立H2O2诱导HepG2细胞氧化损伤模型,采用MTT法检测细胞存活率,流式细胞术检测细胞凋亡。结果组分Ⅰ和模型组的细胞存活率分别为71.9%、50.3%,两组的差异具有显著性(P〈0.05);与模型组比较,组分Ⅰ和Ⅱ均可降低细胞凋亡率(P〈0.05),其中组分Ⅰ更明显。结论高良姜组分Ⅰ和组分Ⅱ均有一定的抗氧化活性,其中组分Ⅰ更明显。

Antioxidant activity of galangal components on HepG2 cells

Abstract:Objective To explore the protection effect of galangal-derived diphenyl heptane(Component I), flavonoids(Component II), volatile oils(Component III) and alcohol extracts(Component IV) on oxidative damage of HepG2 cells. Methods The galangal extracts were separated by vacuum liquid chromatography(VLC), and identified by thin layer chromatography(TLC) and high performance liquid chromatography-mass spectrometry(HPLC-MS). After treated with H2O2, survival and apoptosis of HepG2 cells were determined by MTT and flow cytometry, respectively. Results The survival rate was higher in component I group than that in model group(71.9% vs 50.3%, P〈0. 05). Compared with model group, components I and II reduced the apoptosis(P〈0. 05), especially in component I group. Conclusion The components I and II from galangal possess an antioxidant activity, especially for component I.
