小麦β-1,3-葡聚糖酶性质及其对小麦根腐离蠕孢菌的抑菌作用

摘    要:为了明确小麦根腐离蠕孢菌(Bipolaris sorokinianum)侵染小麦后被诱导表达的β-1,3-葡聚糖酶基因所编码的β-1,3-葡聚糖酶的性质及其与抗病性的关系,采用生物信息学的方法对β-1,3-葡聚糖酶的一级结构、信号肽和跨膜结构进行了预测分析。结果表明β-1,3-葡聚糖酶是一种分泌型碱性蛋白酶,主要存在于胞外。将克隆到的β-1,3-葡聚糖酶基因构建成重组质粒并转化大肠杆菌BL21,经IPTG诱导后,分离纯化得到了原核表达的β-1,3-葡聚糖酶,对其在体外对小麦根腐离蠕孢菌生长的抑制作用的研究发现,β-1,3-葡聚糖酶对小麦根腐离蠕孢具较强抑菌活性。

Abstract:In order to understand properties and disease resistance of Wheat β-1,3-Glucanase coded by β-1,3-Glucanase gene which was induced by Bipolaris sorokinianum. Bioinformatics method was used to predict the primary structure ,signal peptide and trans-membrane structure of β-1,3-Glucanase. The results of the study showed that β-1,3-glucanase is extracellular (secreted) protein, it was alkaline enzyme. Recombinant plasmid of Wheat β-1,3-Glucanase gene was constructed, the recombinant plasmid was transformed into E. coli BL21, and was induced with IPTG to express recombinant Wheat 13-1,3-Glucanase, the expression of prokaryotic expression of β-1,3-Glucanase was received; And in vitro inhibition of Wheat β-1,3-Glucanase to Bipolaris sorokinianum was researched, the results of the study showed that β-1,3-glucanase has active inhibitor to Bipolaris sorokinianum In vitro.
