美洲品系葡萄叶多酚的提取工艺优化及抗氧化活性研究

摘    要:以美洲品系葡萄叶为原料,研究提取液浓度、温度、时间及料液比对多酚提取率的影响,通过正交实验优化工艺条件,并对体外抗氧化效果进行了研究。结果表明:最佳工艺条件为乙醇浓度50%,浸提温度70℃,浸提时间60 min,料液比1∶15,多酚提取率可达1.72%。美洲品系葡萄叶多酚对1,1-二苯基-2-三硝基苯肼(DPPH)自由基、羟基自由基(·OH)和超氧阴离子自由基(O2-·)具有良好的清除能力,其清除率在样品浓度为1.0 mg·m L-1时,分别为L9:87.74%、64.92%和65.00%;L10:86.15%、58.00%和51.20%。

Abstract:American grape leaves were used as raw materials to study the effect of extraction solvent,temperature,time,and solid to liquid ratio on the extraction rate of polyphenols,the process conditions were optimized by orthogonal experiment,and antioxidant activities were tested as well.The results showed that the optimized extraction conditions were as follows:extraction solvent 50%ethanol,temperature 70℃,time of 60 min,solid to liquid ratio 1∶15. Under these conditions,the extraction rate of polyphenols was 1.72%. The polyphenols extraction showed strong scavenging activities on 1,1-diphenyl-2-picrylhydrazyl(DPPH)radical,hydroxyl redical (·OH)and superoxide redical (O2-·)methods,the scavenging rates for the radicals were 87.74%、64.92% and 65.00%(L9);86.15%、58.00%and 51.20%(L10),respectively,when the concentration was 1.0 mg·mL-1.
