青藤碱、吴茱萸碱和川芎嗪抗细菌内毒素作用及其机制研究

摘    要:为探讨3种中药生物碱青藤碱、吴茱萸碱和川芎嗪对细菌内毒素(脂多糖,LPS)的治疗作用,ICR小鼠腹腔注射6 mg.kg-1的内毒素3 h后,根据分组情况分别再腹腔注射高(10 mg.kg-1)、中(5 mg.kg-1)、低(1 mg.kg-1)3个剂量的青藤碱、吴茱萸碱和川芎嗪,并设立LPS攻毒组、空白对照组和药物对照组。记录LPS攻毒后48 h内各组小鼠临床表现,测定体温、体质量及器官指数,检测血清中LPS和炎症因子的含量。结果显示:青藤碱和川芎嗪能显著抑制体温升高,吴茱萸碱能显著抑制体质量下降和血清中LPS水平升高,青藤碱能显著降低血清中炎症因子含量增加,吴茱萸碱和青藤碱能显著改善小肠肿大等病变。结论:青藤碱、吴茱萸碱和川芎嗪3种中药生物碱具有明显的抗细菌内毒素作用。

Abstract:To investigate the antiendotoxin action of sinomenine,evodiamine and chuanxionggzine,mice in treatment groups were challenged with 6 mg·kg-1 lipopoly saccharide(LPS)by peritoneal injection,and then treated with sinomenine,evodiamine and chuanxionggzine at three concentrations(1,5 and 10 mg·kg-1).The mice in LPS group were challenged with 6 mg·kg-1 LPS by peritoneal injection.The mice in drug groups were treated with 10 mg·kg-1 sinomenine,evodiamine and chuanxionggzine by peritoneal injection.The mice in blank group were treated with physiological saline.The determined items and results were as follows:compared with LPS control group,the body temperature significantly decreased in the high-dose group of sinomenine and the middle-and high-dose groups of chuanxionggzine;the body weight significantly increased in the high-dose group of evodiamine;the LPS contents significantly decreased in the middle-and high-dose groups of evodiamine;the tumor necrosis factor alfa(TNF-α) contents significantly decreased in the middle-and high-dose groups of sinomenine;the interleukin 1 beta(IL-1β)contents significantly decreased in the middle-and high-dose groups of sinomenine;the index of intestinal significantly decreased in the high-dose groups of sinomenine and evodiamine.It suggested that sinomenine,evodiamine and chuanxionggzine had significant antiendotoxin effect,thus to providing a theoretical basis for clinical application of alkaloid in controlling the diseases caused by LPS.
