转绵羊IRF-1基因牛胎儿成纤维细胞的BVDV抗性研究

摘    要:为研究IRF-1基因在抗病毒方面的作用,将IRF-1基因转染牛胎儿成纤维细胞,制备了瞬时表达IRF-1基因的转基因细胞,再将携带单股正链RNA的牛病毒性腹泻病毒(BVDV)作用于上述转基因细胞,观察该病毒在转基因细胞和非转基因细胞上的滴度变化及2种细胞形态、增殖、凋亡和基因表达情况上的差异。结果显示:与非转基因细胞相比,病毒在转基因细胞上滴度显著提高,病毒作用48和72 h后转基因细胞病变程度明显低于非转基因细胞;流式细胞仪检测发现72 h时细胞凋亡率显著降低,荧光定量PCR检测结果显示,转基因细胞中IRF-1基因的表达水平显著提高。结论:IRF-1基因具有促进细胞抗BVDV感染的作用。

Abstract:To evaluate the antivirus efficiency of IRF-1,transfected bovine fetal fibroblast(BFF)that transiently expressed IRF-1 were prepared,and then the above cells were treated with bovine viral diarrhea virus(BVDV)which belonged to positive-strand RNA virus.Cells were examined on their TCID50,cell morphology,growth curve,apoptosis and gene expression level.The results showed that compared to non-transfected cells,the TCID50 on the transgenic cells was significantly higher,the cytopathic effect(CPE)of the transgenic cells was remarkably alleviated at 48 and 72 h,and the transfected cells′ apoptosis rate was also significantly lower by the flow cytometric analysis,whereas the expression levels of IRF-1 was remarkable higher in the transgenic cells.The results indicated that IRF-1 played an important role in BVDV control.
