烟草烟碱转化株早期诱导鉴定的有效性研究

摘    要:以白肋烟为材料研究了采用乙烯利处理对烟碱转化株进行早期鉴定的有效性以及取样时间和部位对鉴定效果的影响。结果表明,乙烯利处理早期鉴定的转化株包括了所有调制后自然形成的转化株,还可以鉴别出一些在正常晾制条件下不能有效鉴别的低转化株;早期测定的烟碱转化率一般高于烟叶自然晾制后的烟碱转化率,有利于提高鉴定的准确性和有效性。根据鉴定结果将非转化株、低转化株和高转化株分别自交留种,后代群体分别有8.6%,59.8%和100%的转化株,表明鉴定方法是有效的,且通过转化株的鉴别和清除可以对品种进行有效改良。对转化株的鉴定从苗期到开花前都可以进行,移栽后4~6周取样测定更利于大批量处理和检测。不同部位叶片都可用于早期诱导和鉴定,但下部叶诱导时间短,便于操作。

Abstract:Burley tobacco was used to investigate the availability of early identification of nicotine converters by etheryl treatment,and the effects of sampling time and leaf position on the identification results.The results showed that the early-identified converters by etheryl treatment matched all the converters identified after normal air-curing,and was able to identify some low converters which was hard to be identified normally.The percent nicotine conversion by early identification was higher than that measured after air-curing,so as to increase the accuracy and availability of identification.Based on the early identification, the non-converters,low converters and high converters were self-crossed respectively and seeds were produced.The 3 populations present 8.6%,59.8% and 100% converters,respectively,in next generation,indicating the high effectiveness of the identification method.The variety was able to be improved by identification and removal of converters from the population.The identification process could be carried out from seedling stage to pre-flowering,although sampling at 4-6 weeks after transplanting was easier to handle.Leaves from all positions could be used in the early identification,while the bottom leaves were ideal because of their shorter inducing time.
