谷子籽粒醇溶蛋白提取条件及A-PAGE方法研究

摘    要:在小麦醇溶蛋白电泳方法的基础上研究了适宜谷子醇溶蛋白的提取条件及电泳方法。结果表明:10粒种子加入200μl 50%异丙醇+1%β-巯基乙醇提取液,在60℃水浴下提取30 min可以得到适宜A-PAGE电泳的醇溶蛋白提取液;在凝胶丙烯酰胺浓度为12.5%,电极液含有0.4‰甘氨酸、8‰冰乙酸情况下,可以得到分辨率高,重复性好的醇溶蛋白谱带,可对谷子品种进行有效的鉴定。

Abstract:The extracting condition and electrophoresis method of millet alcohol-soluble protein was studied based on wheat gliadin gel electrophoresis method.The results indicated that the extraction of prolamin was most efficient with extraction solution composed of isopropyl alcohol(50%) and β-mercaptothanol (1%) at the temperature of 60℃ for 30 minutes.The clear and sepereated alcohol-soluble protein bands were obtained with the concentration of acrylamide at 12.5% and the electrode solution composed of 0.4‰ glycin and 8‰ acetic acid glacial.It was a efficient electrophoresis method to identify and evaluate millet varieties.
