摘    要:以濒危药用植物天山雪莲叶片为研究材料进行组织培养和植株再生研究,旨在建立天山雪莲的高效植株再生体系,为其种群复壮、资源保护与可持续利用提供技术参考。研究结果表明:适合天山雪莲叶片愈伤组织诱导的最佳培养基为MS+2,4-D(0.5 mg/L)+6-BA(1.5 mg/L),诱导出愈率为97%;适合愈伤组织丛生芽分化的最佳培养基为MS+NAA(0.05 mg/L)+6-BA(0.4 mg/L)+水解乳蛋白(500 mg/L),分化率为66%,增殖倍数为5.1;适合生根的最佳培养基为1/2 MS+NAA(0.3 mg/L),在此条件下,根发育良好,生根率为76.9%,植株健壮;组培苗炼苗后移栽,成活率可达89%。

Abstract:An efficient micro-propagation system for Saussurea involucrata Kar.et Kir.,an endangered Chinese medicinal plant,has been developed.97% of callus formation frequencies were achieved from leaf explants when cultured on a MS medium containing 0.5 mg / L 2,4-D(2,4-dichlorophenoxyacetic acid) and 1.5 mg / L 6-BA(6-benzyladenine).66% of adventitious bud regeneration frequency and 5.1 buds per explant were achieved from leaf-derived callus as the medium containing 0.05 mg / L NAA,0.4 mg / L 6-BA and 500 mg / L LH(Lactalbumin hydrolyzates).Up to 76.9% of the regenerated shoots formed complete roots on a halfstrength MS medium containing 0.3 mg / L NAA,and 89% of the complete plantlets survived and grew vigorously under greenhouse condition.
