大孔树脂纯化马尾松花粉总黄酮的工艺研究

摘    要:以马尾松花粉为材料,筛选对马尾松花粉总黄酮吸附和解吸性能好的大孔树脂,并对大孔树脂纯化马尾松花粉总黄酮的工艺条件进行优化。结果表明,弱极性的DM-130大孔树脂对马尾松花粉总黄酮具有较好的吸附与解吸性能,其吸附率和解吸率分别达到84.24%和93.32%。DM-130大孔树脂纯化马尾松花粉总黄酮的最佳工艺参数为:上样质量浓度0.68 mg/m L,上样流速0.67 m L/min,上样体积为5倍柱床体积,洗脱溶剂为75%乙醇,洗脱流速0.67 m L/min,洗脱剂用量为6倍柱床体积。纯化后马尾松花粉总黄酮的纯度可达到50.4%。

Purification of Total Flavonoid from Pinus massoniana Pollen with Macroporous Resin
Abstract:Using the Pinus massoniana pollen as the raw material, the suitable macroporous resin type with good adsorption and desorption capacity for total flavonoid from Pinus massoniana pollen was screened, and the purification conditions were optimized. The results showed that, DM-130 macroporous resin with weak polarity had higher adsorption and desorption rate to total flavonoid, which were 84.24% and 93.32%, respectively. The optimal purification conditions were as follows: the sample concentration 0.68 mg/mL, sample flow rate 0.67 mL/min, sample volume 5 BV (bed volume), 75% ethanol as eluent, elution flow rate 0.67 mL/min, and the eluent volume 6 BV (bed volume). The purity of total flavonoid from Pinus massoniana pollen after purification could reach 50.4%.
