摘    要:建立了同时测定花生壳中绿原酸、3,4-二咖啡酰奎尼酸和木犀草素3种有效成分含量的反相高效液相色谱方法。采用Kromasil C18柱(200 mm×4.6 mm,5μm),乙腈和0.1%甲酸水溶液梯度洗脱,流速为1.0 mL/min,检测波长340 nm,柱温27℃。在该色谱条件下,绿原酸、3,4-二咖啡酰奎尼酸和木犀草素的质量浓度分别为3.75~60μg/mL(R2=0.999 9),3.63~58.08μg/mL(R2=0.999 9),17.25~276μg/mL(R2=0.999 7)内与色谱峰面积呈现良好的线性关系,平均加标回收率(n=5)为121.6%,97.79%,124.35%,RSD分别为0.71%,1.49%,0.44%。该分析方法快速准确、重现性好,可应用于花生壳资源产业开发中质量标准的建立和完善。

Abstract:A method of reversed-phase high performance liquid chromatography is established for the simultaneous determination of chlorogenic acid,3,4-O-dicaffeoylquinic acid and luteolin in peanut hull.Separation is achieved using a Kromasil C18 column(4.6 mm × 200 mm,5 μm),under the gradient elution with the mobile phase of acetonitrile and 0.1% formic acid / water,flow rate is 1.0 mL/min,detected at 340 nm and under 27 ℃.The linear ranges of 3 components are 3.75~ 60 μg/mL(R2=0.999 9),3.63 ~58.08 μg/mL(R2=0.999 9) and 17.25 ~ 276 μg/mL(R2=0.999 7) respectively.The average recoverises(n=5) are 121.60%,97.79%,124.35% and with RSD of 0.71%,1.49%,0.44% respectively.The method is simple,rapid and highly sensitive,suitable for the quality control of peanut hull.
