聚甘露糖醛酸和聚古罗糖醛酸的制备与鉴定

摘    要:褐藻胶经过稀HCl降解,pH值2.85分级获得2种聚糖粗品。通过dowex阴离子交换分离,浓度0.5~2.0 mol/L的醋酸梯度洗脱提纯,即获得高纯度的聚甘露糖醛酸(PM)和聚古罗糖醛酸(PG)。采用高效液相色谱技术对其分子量及纯度进行初步测定,利用核磁共振技术对其分子结构和纯度进行最终鉴定。由核磁共振图谱可得其结构保持了M,G的原有结构,且制得的2种聚糖纯度均在95%以上。

The Preparation and Identification of Polymannuronic Acids and Polyguluronic Acids
Abstract:Algin can be classified into two crude samples of glycan successively after degrading by dilute hydrochloric acid and grading in pH 2.85.The polymannuronic acid(PM)and polyguluronic acid(PG)in high purity can be purified by the two crude samples of glycan through dowex anion exchange separation by gradient elution of 0.5~2.0 mol/L acetic acid.PM and PG's molecular weights and purities are primarily detected by the high performance liquid chromatography technology.The final identification of PM and PG's molecular structure and purities use the NMR technique.This separation method can keep the original structures of M and G,whose purities are more than 95%.
