摘    要:采用AFLP技术,用混合的模板DNA从数对引物中筛选出6对带型分布均匀、多态性丰富且分辨能力强的引物,对市场上经营的两份大白菜杂交种样品586和587所扩增出的AFLP指纹图谱进行分析,根据数据分析得到了两份大白菜样品与标准样品之间的欧氏距离系数。6对引物对两份大白菜样品和标准样品扩增所得到的指纹图谱分析的结果表明,587与标准样品存在显著的差异,其中多态性位点占总扩增位点的31.7%,欧氏距离系数为3.87,样品587与标准样品不是同一个品种;而样品586与标准样品之间欧氏距离系数近于0,而据此得知此样品与标准样品之间没有明显的差异,判断为同一个品种。本研究采用AFLP技术很好地鉴别了两份样品的不同。这对应用AFLP技术鉴定大白菜品种的真伪,保护品种繁育者、生产者和种子使用者的利益提供了有效技术体系的依据。

Abstract:Six pairs of AFLP primers from many pairs have been selected, which had rich polymorphism. The genetic diversity of two Chinese cabbage samples and standard sample were analyzed with the fingerprinting amplified by 6 pairs of primers, and Euclidean distances coefficient were estimated. The results showed that there were distinct differences between the sample 587 and standard sample, and the rate of the polymorphism bands was 31.7%, which demonstrated that 587 was a different variety from the standard sample; there were no distinct differences between sample 586 and standard sample, it showed that 586 was the same variety as the standard sample. This indicated that two Chinese cabbage samples could be identificated clearly by using AFLP. AFLP fingerprinting provides the best currently available technology to establish differentiation for plant variety identification.
