厚皮甜瓜品种组合SSR指纹图谱构建

摘    要:构建厚皮甜瓜品种DNA指纹图谱,为厚皮甜瓜品种DUS测试和品种真实性鉴定技术奠定基础。利用SSR标记技术对21份厚皮甜瓜品种组合进行DNA指纹分析,筛选合适的多态性引物,初步建立其SSR指纹图谱。从700对SSR引物中筛选23对多态引物组合,对所有供试材料进行分析,共检测出63条多态性条带,平均每个引物多态性条带为2.74条;平均多态性信息量(PIC)为0.43,变化范围为0.28~0.69;应用其中的6对引物组合(CMGAN12、MU7161、F22_85、F21_16、ECM79和MU5499)获得了每个品种组合的SSR特征带,初步建立21份厚皮甜瓜品种组合的SSR指纹图谱,可以将所有供试材料区分。SSR标记技术适于构建厚皮甜瓜品种DNA指纹图谱;为品种的真伪鉴定和知识产权保护提供有效的科学依据。

Abstract:The aims of this study were to construct a DNA fingerprinting of varieties ofCucumis melo ssp. melo ,which could provide a possibility of establishment of DUS and accession identification ofCucumis melo ssp.melo. Simple sequence repeat (SSR) markers were used to analyze the DNA fingerprinting of 21 varieties and then to choose suitable primers for the construction of DNA fingerprinting ofCucumis melo ssp.Melo. 23 pairs were screened out of 700 SSR primer pairs with polymorphism among 21 varieties and 63 polymorphic bands were obtained. 2.74 polymorphic bands in average were amplified by a pair of SSR primers. The polymorphic information content values (PIC ) ranged from 0.28 to 0.69,with a mean of 0.43. 6 SSR primer pairs,with high polymorphism and good repeatability,which could identify 21 varieties,were selected from 23 SSR primer pairs. As a result,a total of 21 varieties could be identified by the 6 SSR primers. Finally,a DNA fingerprinting containing 21 varieties was constructed by using those 6 SSR primers. The results indicated that SSR markers were suitable for the construction of DNA fingerprinting ofCucumis mel o ssp.melo. Moreover,the fingerprinting will provide reliable scientific evidence for the protection of intellectual property right for these varieties.
