摘    要:为研究高渗溶液对猪肺炎支原体(Mhp)形态、大小,以及活力的影响。Mhp经终浓度为1.8% NaCl的高渗溶液分别作用1、2、4、6、8 min后,分别进行染色镜检、粒径分析仪测定粒径、CCU测定和PCR鉴定。结果表明:在高渗溶液作用不同时间后,从2 min开始,Mhp皱缩变圆,且直径变小,菌体数量也快速变少至消失;测定的终CCU降低了一个梯度,且时间延长了1天;在作用不同时间后的样品中,PCR均能扩增出Mhp目的条带。经终浓度为1.8% NaCl作用1~6 min,Mhp的形态大小发生了变化,且具有活性,这将为Mhp经肌肉注射进入血液循环研究和猪支原体肺炎活疫苗的新免疫途径研究提供参考数据。

Abstract:The present study was designed to investigate the effect of hypertonic solution on the morphology, size, and dynamic of Mycoplasma hyopneumoniae (Mhp). With a final concentration of 1.8% NaCl, Mhp was stimulated for 1, 2, 4, 6, 8 min. Then these samples were analyzed by dyeing microscopic examination, particle size analyzer, CCU determination and PCR, respectively. The results showed that the morphology and size of Mhp had changed, such as shrinkage and round, smaller diameter, number of bacteria decreases rapidly, or even disappear, after which Mhp was effected for 2 min. CCU was reduced, and observation time was extended. The target band could be amplified for samples of Mhp. Therefore, it was concluded that the morphology and size had changed when Mhp were stimulated with a final concentration of 1.8% NaCl for 1-6 min, but they also have activity. Moreover, this study would also provided reference data for studing a new immunization route of Swine Mycoplasma Pneumonia vaccine and the blood circulation for Mhp by intramuscular injection.
