紫花鸢尾花粉萌发及贮藏性研究

摘    要:以紫花鸢尾的花粉为试材,运用正交试验设计方法,研究了不同培养基对其萌发的作用,同时探讨不同贮藏条件和贮藏时间对花粉生活力的影响。结果表明:(1)紫花鸢尾花粉在200 g/L蔗糖+ 300 mg/L H3BO3 + 200~300 mg/L CaCl2的液体培养基上萌发率最高,为81.55%。(2)干燥条件有利于紫花鸢尾花粉的贮藏,且硅胶为适宜的干燥剂。(3)随着贮藏时间的延长,紫花鸢尾花粉生活力下降,但随着贮藏温度的降低,花粉生活力下降速度变缓,在-196℃下保存270天后的花粉萌发率为84.21%,与新鲜花粉生活力差异不显著。

Abstract:The effect of different media on pollen germination of Iris ensata through orthogonal experiment, in other hand, the effects of storage condition, storage temperature and storage time on pollen germination were studied. The results showed that: (1) the optimum medium was sucrose 200 g/L + H3BO3 300 mg/L + CaCl2 200-300 mg/L, in which the rate of germination was 81.55%. (2) Dry condition was suitable for pollen storage of Iris ensata, and the silica gel was the suitable desiccant. (3) The pollen viability decreased over time, but the viability decreased slowly with the falling of temperature, pollen stored at -196℃ for 270 days, its germination rate was 84.21%, which was not significant with fresh pollen.
