从核糖体DNA ITS区序列研究甘蔗属及其近缘属种的系统发育关系

摘    要:测定了甘蔗属及其近缘属的13个种共43个个体和1个橡草(Pennisetum schumach)外群的核糖体DNA中的内转录间隔区(ITS)及5.8S rDNA 基因的序列.结果表明:甘蔗属及其近缘属种的ITS区(含ITS1,5.8S rDNA和ITS2)序列的长度范围为589~591bp,变异位点为140个,信息位点为60个;其中,ITS1和ITS2的长度范围分别为205~208bp和216~220bp,

Abstract:The internal transcribed spacer(ITS) regions of nuclear ribosomal DNA from 14 species of Saccharum and related genera were sequenced. The regions ranged in length from 589 to 591bp, with ITS1(205~208bp)slightly shorter than ITS2(216~220bp). Of the 591bp aligned positions, there were 140 variable sites(64 in ITS1, 68 in ITS2, 8 in 5.8S region), and 60 informative sites(33 in ITS1, 24 in ITS2, 3 in 5.8S region). The neighbor joining tree based on the ITS sequences analysis with P. schumach as outgroup indicated that Zhejiang fruit sugarcane should belong to S. officinarum. N. fallax and E. fulvus, N. porphyrocoma and E. rockii should be placed in a same genus. Arundinaceum has different variable sites from other species, it is an independent clade isolated from Saccharum and Erianthus . The present analysis demonstrated that Arundinaceum should not belong to Saccharum , which is consistent with the results from morphology and cytology.
