体外免疫法制备单克隆抗体的研究

摘    要:本研究进行了三次体外免疫制备单克隆抗体(McAb)的试验,三次融合率分别为:85.59%(493/576),84.55%(487/576)和85.07%(490/576),而体内免疫法为87.33%(503/576)。在三次试验中,有两次产生抗体,经克隆后,与55匹含有靶抗原的马红细胞反应中,各次试验获得的各株McAb的最低反应率分别为85.45%(47/55),80.00%(44/55),而体内免疫法为83.60%(46/55)。上述结果表明:小鼠脾细胞在体外培养4d后,并不影响其与SP2/0(小鼠骨髓瘤细胞)的融合,而且在体外可接受抗原的刺激引起免疫应答,并产生抗体,其特异性与体内免疫法相似。

Abstract:Three times test in vitro immunization were performed for production monoclonal antibodies (McAb). The result showed that fusion rates were 85. 59% (493/576);84. 55% (487/576) and 85. 07% (490/576) respectively, but it was 87. 33% (503/576) in vivo immunization (control). Among three tests,two were successful for antibodies production. After having been cloned, the antibody specificity was detected by using red cells from 55 horse, which contained target antigen.The result showed that positive rate at least were 85.45% (47/55) and 80. 00% (44/55) respectively,but it was 83. 60% (46/55) in the control.The result indicated that after mice spleen cells had been cultured for four days,it fused with SP2/0 was still effective, and the spleen cells could responsed to antigen externally and produced antibodies.The antibodies specificities were similar to the control.
