杜仲叶绿原酸的微波提取及高速逆流色谱分离纯化

摘    要:以杜仲叶为原料,正交试验优化了微波辅助提取绿原酸的工艺条件。应用高速逆流色谱(HSCCC)分离制备高纯度杜仲叶绿原酸,通过质谱技术与核磁共振技术进行结构鉴定。结果表明,杜仲叶最佳提取工艺条件为微波辅助50%的丙酮水溶液浸提,料液比1∶30,提取时间25min,提取功率400W,绿原酸的提取率达2.28%;高速逆流色谱法...

Abstract:The optimum extraction of chlorogenic acid from Eucommia ulmoides leaves using microwave-assisted solvent extraction was studied by orthogonal experiments. The chlorogenic acid was extracted by high-speed counter-current chromatography (HSCCC), and the structure of chlorogenic acid was identified by Mass Spectrometry (MS) and Nuclear Magnetic Resonance (NMR) technologies. The results showed that the optimized extracting conditions of chlorogenic acid was 50% acetone, extraction temperature 50℃, material to solvent 1 ∶30, extraction time 25min, microwave power 400W, the extracting rate of chlorogenic acid could reach 2.28%. The best condition of HSCCC was ethyl acetate/n-butanol/water=3/1/4 as solvent system; photogenic as the stationary phase, phasor as the mobile phase; speed 900r/min; flow rate 2.0ml/min; constant temperature 30℃ and 98.7% purity of chlorogenic acid was separated and purified only though one step method.
