摘    要:为了提高玉米直链淀粉含量,用基因枪将前期工作中构建的sbeⅡb RNAi表达载体pBAC413导入玉米自交系幼胚愈伤组织,经过筛选获得了12 株转化再生植株,其中6 个植株获得了结实种子。DNA点杂交、PCR扩增和PCR-Southern均证明目的基因已整合到T0代再生植株玉米基因组中。对T1代转基因玉米籽粒淀粉含量进行测定,结果表明总淀粉含量比对照没有显著变化,其中1个转基因玉米株系的直链淀粉含量比对照提高了15.6%。

Abstract:To regulate the biosynthesis of maize amylose, an expression vector of RNAi with the fragment of sense gene +FAD2 intron +antisense gene had been constructed, and. was transformed to maize calli which were induced to form from the elite inbred lines by Biolistic PDS1000/He. The selection was carried out on the media containing concentrations of xylose. 12 transformed plantlets were obtained. The integration of alien gene was conformed in 6 transformed plantlets with the analysis of DNA dot blotting, PCR and PCR-Southern hybridization. The seeds amylose content of regeneration plants was detected,and amylose content of a transgenic line increased up to 15.6%.
