摘    要:以积雪草(Centella asiatica)的春芽作为外植体,以MS培养基为基本培养基,添加不同激素,分别组配成12种愈伤组织诱导培养基和丛芽增殖培养基进行组织培养,然后将诱导的积雪草丛芽切成单株后接入4种生根培养基,研究不同激素配比对愈伤组织的形成、丛芽的增殖及生根的影响.结果表明积雪草春芽在MS+6-BA 1.0 mg/L+NAA 0.20 mg/L+2,4-D 0.5 mg/L培养基上容易诱导愈伤组织,诱导率为86%.丛芽在MS+6-BA 3.0 mg/L+NAA 1.0 mg/L+活性炭0.1 g/L培养基上的成活率为94%,增殖系数为5.9.以MS+6-BA 0.4 mg/L+NAA 0.2 mg/L+活性炭0.1 g/L为培养基较有利于生根培养.

Abstract:The spring bud of Centella asiatica was used as explants and MS as basic medium,12,12 and 4 mediums for induction,differentiation and rooting of Centella asiatica were screened to find the best medium respectivily.The effects of different combinations of hormone on the induction,differentiation and rooting of callus were studied.The results showed that the callus of C.asiatica spring bud could easily be induced on the medium of MS+6-BA 1.0 mg/L+ NAA 0.20 mg/L+ 2,4-D 0.5 mg/L,with an induction ratio of 86%.The survival rate of clumpy buds was 94% and the multiplication coefficient was 5.9 on the medium of MS+6-BA 3.0 mg/L+NAA 1.0 mg/L+Activated carbon 0.1 g/L.The optimum medium for rooting induction was MS+6-BA 0.4 mg/L+NAA 0.2 mg/L+Activated carbon 0.1 g/L.
