不同脱毒技术对马铃薯病毒脱毒效果的影响

摘    要:以三个感染了不同病毒的马铃薯品种作为试验材料,研究了3种不同的马铃薯脱毒技术,对脱毒效率及脱毒苗的成活率进行对比研究,筛选出不同病毒种类最适宜的脱毒技术方法。用茎尖剥离,热处理结合茎尖剥离技术和病毒唑结合茎尖剥离技术,设计不同的热处理周期和病毒唑浓度对材料进行脱毒处理,在计算植株的成活率及成苗率的基础上对其进ELISA检测法检测脱毒率。试验结果表明以上三种方法中热处理加茎尖剥离和病毒唑加茎尖剥离的复合脱毒的方法在提高试管苗的脱毒率上有明显的优势,单一的茎尖剥离处理的茎尖成活率及脱毒率普遍较低。通过变温热处理加茎尖剥离处理中(5周)热处理时PVY, PLRV, PVS均可达到较高的脱毒。脱毒率最高为V病毒100%。病毒唑浓度为45mg/L时,通过病毒唑及茎尖剥离方法得到的PVY和PLRV的最高脱毒率为100%。PVS的最高脱毒率为92%(结论)。比起单一的茎尖剥离技术复合脱毒技术较有优势,在5周的热处理加茎尖剥离技术(只取一个叶原基的茎尖)处理适合PVS病毒的脱毒。变温热处理培养后茎尖剥离处理适于PVY病毒脱毒。试验中三种方法均适合脱去PLRV病毒。经过设计不同的热培养温度和化学处理浓度...

Abstract:In this paper, three potato varieties infected with different viruses were used as materials to study three different virus-free technologies for potato, and the virus-free efficiency and the survival rate of virus-free seedlings were compared to screen out the most suitable virus-free technology for different virus types.The technology of stem tip stripping, heat treatment combined with stem tip stripping and ribavirin combined with stem tip stripping were used. Different heat treatment cycles and ribavirin concentrations were designed to detoxify the materials. On the basis of calculation of the survival rate and seedling rate, the virus-free efficiency was detected by ELISA. Among the above three methods, the combined detoxification method of heat treatment plus stem tip stripping and ribavirin plus stem tip stripping had obvious advantages in improving the virus-free efficiency of test-tube plantlets. The survival rate and virus-free efficiency of the stem tip of a single stem tip stripping treatment were generally lower. PVY, PLRV and PVS could achieve higher virus-free potato during heat treatment (5 weeks) in variable temperature heat treatments and stem tip stripping treatment. The highest detoxification rate was 100% for V virus. When the concentration of ribavirin was 45mg/L, the highest virus-free efficiency of PVY and PLRV obtained by ribavirin and stem tip stripping were 100%. The highest detoxification rate of PVS was 92%. In conclusion,compared with the single stem tip stripping technology, the combined virus-free technology has advantage, and the treatment of 5 weeks of heat treatment plus stem tip stripping technology (only taking the stem tip of one leaf primordium) is suitable for PVS virus-free. Stem tip stripping treatment after variable temperature heat treatment culture is suitable for PVY virus detoxification. The three methods are all suitable for removing PLRV virus. After different thermal culture temperatures and chemical treatment concentrations, the virus-free efficiency of different potato viruses can be effectively improved.
