“晶瑶”草莓叶片再生体系的建立

摘    要:以草莓"晶瑶"为试材,研究了暗培养时间、培养温度、激素配比、硝酸银浓度、不同植物凝胶等对不定芽再生的影响,建立了"晶瑶"草莓叶片的离体再生体系。结果表明:硝酸银对"晶瑶"草莓叶片再生有抑制作用;暗培养15 d、28℃的培养温度、以Phytagel为凝固剂有利于"晶瑶"草莓叶片再生;TDZ 1.5 mg/L+IBA 0.3 mg/L是较好的诱导培养基激素配比,此时不定芽的再生率达到76.77%,平均再生芽数达到4.3个。

Abstract:In this study,the effects of dark time,temperature,AgNO3 concentration,plant growth regulator and plant gel on the regeneration of adventitious bud from leaf explant of strawberry "Jingyao" were investigated,and an efficient leaf regeneration system was constructed.The results showed that the addition of AgNO3 to the medium had inhibitory effect on the regeneration of adventitious bud from leaf;the following cultivation conditions were in favor of the regeneration of adventitious bud from leaf: 15 days' dark treatment,temperature 28 ℃ and coagulator 2.5 g/L Phytagel;the optimum medium for adventitious bud regeneration was MS supplemented with 1.5 mg/L thidiazuron(TDZ) and 0.3 mg/L indole-3-butyric acid(IBA).Under the above-mentioned optimum conditions,the isolated regeneration rate of adventitious bud from leaf explant reached 76.77%,and the average number of regenerated buds reached 4.3.
