植物激素对玉米愈伤组织诱导的影响

摘    要:设计了含有不同植物激素的4种培养基用于玉米未成熟合子胚愈伤组织的诱导,以确定植物激素在玉米愈伤组织诱导中的作用。结果表明,KT和6-BA会明显降低愈伤组织和胚性愈伤组织的诱导频率,对愈伤组织的生长有明显的抑制作用;2,4-D在愈伤组织诱导中起着重要作用,但是在诱导培养基中添加3.3mg/L Dicamba比2 mg/L 2,4-D的诱导效果更好,除了能提高愈伤组织诱导频率外,更重要的是能提高胚性愈伤组织比例和获得更好的愈伤组织生长状态;并获得了一个用于从玉米未成熟合子胚高效诱导出良好愈伤组织的培养基配方LM6-CI。

Abstract:In this study, four culture media, including 2,4-D, Dicamba, 6-BA and KT were designed and used for maize callus induction to evaluate their functions in callus induction from immature zygotic embryos. As a result, 6-BA and KT, added in induction media, decreased calli initiation frequency and embryonic calli initiation percentage, and their growth rate had been strained; 2,4-D played an important role in callus induction, but, 3.3 mg/L Dicamber was better than 2 mg/L 2,4-D, with higher calli induction frequency and embryonic callus percentage. An elite media compositions used for callus induction from maize immature zygotic embryos was obtained successfully in this study.
